    • High-throughput sequencing of Arabidopsis microRNAs: evidence for frequent birth and death of MIRNA genes
    • Together with references [7,8] this paper used high-throughput sequencing to identify numerous non-conserved miRNAs and a handful of conserved miRNAs that had escaped previous detection owing to low accumulation levels. Target predictions and validations expanded miRNA regulation to a variety of new gene networks. This paper, together with reference [8], also identified scores of endogenous siRNAs.

    • This paper together with references [25,26] identified that several AGO proteins select their associated small RNAs on the basis of 5′-terminal identity. These results reveal an additional parameter that dictates small RNA guide strand selection, which has implications in artificial miRNA and tasiRNA design.

    • Arabidopsis ARGONAUTE1 is an RNA Slicer that selectively recruits microRNAs and short interfering RNAs
    • This paper together with references [30,33] demonstrates AGO1-mediated cleavage activity of complementary target RNAs, and shows that the DDH motif of the AGO1 PIWI domain is necessary for this activity. They also demonstrate that AGO1 is physically associated with miRNAs, tasiRNAs, and transgene-derived siRNAs, but not 24-nt siRNAs that mediate chromatin silencing. Reference [30] also shows that AGO4 primarily binds to transposon- and repetitive element-derived 24-nt small RNAs and can cleave target RNA transcripts.

    • Target mimicry provides a new mechanism for regulation of microRNA activity
    • This paper identified miRNA target mimicry as an endogenous mechanism that regulates miRNA availability by binding and sequestrating miRNAs. This work also showed the utility of artificial target mimics to regulate the availability of desired miRNA families, providing a platform to analyze partial-loss-of-miRNA function in plants.
