An extensive mathematical and experimental analysis of three potential sources of ultrasensitivity in the inactivation of the Wee1 kinase during mitotic entrance in the Xenopus extracts: zero-order and multisite phosphorylation (intrinsic) and substrate competition (extrinsic). Although multisite phosphorylation contributes some of the sigmoidal response of Wee1 phosphorylation to CDK1 activity in a purified in vitro system, the competition with additional CDK1 targets in these reactions restores the steepness of the response that is found in complete Xenopus egg extracts.
