This paper describes a careful study of tryptophan tag and spacer effects on protein partitioning in hydrophobically modified EOPO/starch systems. Tag exposure is the key to good partitioning.

Here the twist is that the authors weight the scales with the fraction of the protein surface occupied by each amino acid type for each protein. The predictive ability for partition coefficients is good. And, this approach should have implications for predicting solubility in salting-out processes and retention in hydrophobic chromatography.

This is reminiscent of earlier work by Horvath correlating protein solubility in salting-out systems with retention in reversed-phase chromatography. Perhaps HIC can serve as a rapid screening platform to map out partitioning behavior.

This results in repeated precipitation and dissolution, with substances eluting from the centrifuge in order of most soluble to least soluble. CPC sounds more complicated than it actually is and reported separations for several different crude mixtures are impressive.

The performance of this system in terms of PEG-phase yield of enzyme was indistinguishable from that of separate disruption and extraction steps. However, a complicating factor in this approach is that the partitioning behavior of the cell debris in some cases was opposite to that of the separate unit operations. A logical integration somewhat complicated by the vagaries of cell debris partitioning.

Vogel and co-workers paid particular attention to the fluid mechanics of the filter device, using a cone-and-plate geometry to generate a uniform filter surface shear rate - motivating the 'CSAF' moniker - that produces sharp breakthrough curves due to more homogeneous transmembrane flow; the sharper the breakthrough curve, the greater the dynamic binding capacity. The cone-and-plate design is particularly clever in its simplicity and effectiveness.
