Microbial exposure of rural school children, as assessed by levels of N-acetyl-muramic acid in mattress dust, and its association with respiratory health

J Allergy Clin Immunol 2004; 113:860-867. This important article demonstrates that high levels of exposure to peptidoglycan in early life reduces the risk of asthma symptoms. This study indicated that both endotoxin and peptidoglycan share this feature. It illustrates the importance of interacting effects of components of bioaerosols.

Michel O, LeVan TD, Stern D, et al. Systemic responsiveness to lipopolysaccharide and polymorphisms in the toll-like receptor 4 gene in human beings. J Allergy Clin Immunol 2003; 112:923-929. This extends the previous findings of Arbour et al. [39] that polymorphisms in the TLR4 gene result in blunted responses to inhaled endotoxin. In this case, systemic inflammatory responses are diminished in individuals with polymorphic TLR4.

Differential modulation of endotoxin responsiveness by human caspase-12 polymorphisms
-
Saleh M, Vaillancourt JP, Graham RK, et al. Differential modulation of endotoxin responsiveness by human caspase-12 polymorphisms. Nature 2004; 429:75-79. These experiments identify a downstream component of lipopolysaccharide signaling that causes diminished inflammatory response of peripheral blood cells after endotoxin stimulation. This work is important because it demonstrates that polymorphisms in several genes of the lipopolysaccharide signaling pathway may result in altered responses to endotoxin.
