This study demonstrates that the combined treatment of rats maintained on a low-salt diet with captopril and losartan reduced intrarenal angiotensin II levels to a greater extent than treatment with either agent alone. The synergistic actions of these two agents may have important implications for long-term organ protection in hypertension and renal disease.

This paper provides further data supporting enhanced intrarenal angiotensin II in non-clipped as well as in clipped kidneys. The elevated angiotensin II in clipped kidneys was suppressed by a chymase inhibitor but not by an ACE inhibitor, whereas an ACE inhibitor, but not a chymase inhibitor, inhibited the intrarenal angiotensin II levels in the non-clipped kidneys. The data indicate that chymase activity is enhanced in clipped kidneys, whereas ACE-mediated angiotensin II generation is primarily responsible for elevated angiotensin II contents in nonclipped kidneys.

The inappropriate maintenance of nanomolar concentrations of angiotensin II in proximal tubular fluid may contribute to the augmented sodium reabsorption and impaired pressure natriuresis, leading to hypertension.

This paper shows that angiotensinogen mRNA expression in rat proximal tubular cells is increased by high glucose, but this effect is prevented by the administration of an antioxidant and a p38 MAPK inhibitor. The data indicate that the stimulatory action of high glucose on angiotensinogen gene expression in the proximal tubular cells is mediated via reactive oxygen species generation and subsequent p38 MAPK activation.

This paper shows that intrarenal angiotensinogen expression and the urinary excretion of angiotensinogen are significantly increased in angiotensin II-dependent, but not in angiotensin II-independent hypertensive rats, indicating a positive feedback system that leads to the further intrarenal production of angiotensin II during the development of angiotensin II-dependent hypertension.

The administration of a β-adrenoceptor agonist stimulated both renin and renal angiotensinogen expression, providing evidence that catecholamines are involved in the mechanism that alters the expression of renal RAS genes.

Using isolated perfused cortical collecting duct segments, angiotensin II added to the lumen increased intracellular sodium uptake and this effect was prevented after the blockade of the amiloride-sensitlve sodium channel. The results indicate that distal nephron angiotensin II may regulate amiloride-sensitive sodium channel activity.

This study reports that angiotensin I added to the perfusate of cortical collecting ducts leads to angiotensin II stimulation of amiloride-sensitive sodium channel activity and that ACE was present in the cortical collecting duct, supporting the hypothesis that angiotensin I to angiotensin II conversion can occur in the cortical collecting duct.
