Another key signaling pathway which can be activated by IGF-1 and IGF-2 is the Wnt pathway, which regulates the key transcription factor, β-catenin. Signaling though IGF-1R (via PI3K/AKT) can activate Wnt signaling through GSK-3β inhibition. GSK-3β is a key component in the degradation of β-catenin, and therefore in the maintenance of the ‘normal’ state. See M. Illyas J. Pathol. 205 2005 130

There is evidence that antidiabetic drugs, such as the PPARγ agonist thiazolidinedione, rosiglitazone, suppress IGF-1 tumor-promoting activity. See G. He Y.M. Sung J. DiGiovanni S.M. Fischer Cancer Res. 66 2006 1873
