    • Briefly, diluted whole blood was layered on top of an equal of a sucrose density gradient (Histopaque 1077, number 10771, 8, centrifuged, and then the leukocyte layer was collected, washed twice, and resuspended in RPMI 1640. Cells were then plated in 96-well plates (1 × 106 cells/well; n, 4 replicate wells/sample) in RPMI 1640, 5% autologous serum. After 2 h of incubation (41°C, 5% CO2, cells were washed, and media was replaced. After 10 h of incubation, cells were treated with either 0 or 1 μg of lipopolysaccharide (LPS)/mL of media (L7261, Sigma Aldrich [8, n, 2 replicate wells/sample × LPS treatment, At 24 h post-LPS treatment, supernatant was removed, and NO was measured using the Griess method (G4410, Sigma Aldrich [8, as previously described [10, Finally, HD11 cells (avian macrophage-like cell line described by Beug et al, 11, 1 × 10 6 cells/well in a 96-well plate) were cultured with RPMI 1640 plus 5
