After 25-min incubation at 37 °C the reaction was excited at 485 nm and emission was observed at 595 nm in a microplate reader. Positive control reactions were performed in the absence of any inhibitor to simulate an unimpeded ATPase reaction and negative control reactions lacked RecA to simulate the complete inhibition of ATP hydrolysis.
