    • Using cultured embryonic rat spinal commissural neurons, the authors demonstrate that substrate bound netrin-1 triggers growth cone membrane extension and that DCC mediates adhesion to a substrate of immobilized netrin-1. Additionally, the results identify a signal transduction complex recruited to the DCC intracellular domain that is required for the growth cone response to netrin-1.

    • FAK and Src kinases are required for netrin-induced tyrosine phosphorylation of UNC5
    • In addition to the reports indicating that FAK and src kinases are involved in DCC mediated chemoattraction [30-33], this study provides evidence that FAK and Src kinases also play a role in signaling by UNC5 proteins, the receptors that mediate chemorepellent responses to netrin. When expressed with an UNC5 protein, DCC can also contribute to chemorepulsion. Interestingly, this study provides evidence that DCC is not required for, but facilitates tyrosine phosphorylation of the intracellular domain of UNC5B.
