Laboratory in-vitro study examining protective effect of GLN in heat shock factor-1 (HSF-1) knock out cells. GLN led to significant protection against heat injury in HSF-1+/+ cells, but no protection from GLN was observed in HSF-1-/- cells. GLN led to enhanced HSF-1 promoter binding and HSF-1 phosphorylation following stress in HSF-1+/+ cells. GLN also led to enhanced translocation of unphosphorylated HSF-1 in the nucleus prior to stress. Data demonstrates that GLN's protection is dependent on the presence of HSF-1 and GLN can directly activate HSP transcriptional pathway prior to and following heat stress injury.

This study demonstrated the first non-toxic, clinically relevant enhancer of HSP-70 expression in critically ill patients.
