Pharmacological activation of liver X receptors promotes reverse cholesterol transport in vivo
-
Naik SU, Wang X, Da Silva JS, et al. Pharmacological activation of liver X receptors promotes reverse cholesterol transport in vivo. Circulation 2006; 113:90-97. For the first time these authors provide definitive evidence that activation of LXR promotes reverse cholesterol transport form macrophages into the liver, bile, and feces. These data suggest a mechanism by which LXR agonists exert their atheroprotective role.

ABCG1 has a critical role in mediating cholesterol efflux to HDL and preventing cellular lipid accumulation
-
-/- mice accumulate massive amounts of lipid in the lungs and liver, whereas overexpression of ABCG1 has a protective effect. In addition, loss of ABCG1 impairs the ability of macrophages to efflux cholesterol to HDL. Together, the results shown in this paper point to a critical role for ABCG1 in controlling cellular sterol homeostasis.

ABCG1 redistributes cell cholesterol to domains removable by high density lipoprotein but not by lipid-depleted apolipoproteins
-
Vaughan AM, Oram JF. ABCG1 redistributes cell cholesterol to domains removable by high density lipoprotein but not by lipid-depleted apolipoproteins. J Biol Chem 2005; 280:30150-30157. Overexpression of ABCG1 results in increased levels of cholesterol in the plasma membrane and increased cholesterol efflux to HDL. In addition, biotinylation of cell surface proteins suggests that some ABCG1 protein resides in the plasma membrane.

ABCA1 and ABCG1 synergize to mediate cholesterol export to apoA-I
-
Gelissen IC, Harris M, Rye KA, et al. ABCA1 and ABCG1 synergize to mediate cholesterol export to apoA-I. Arterioscler Thromb Vasc Biol 2006; 26:534-540. Overexpression of ABCG1 promotes increased cholesterol efflux to HDL, phosphatidylcholine vesicles, and conditioned media from ABCA1-overexpressing cells. The authors suggest a cooperative interaction between ABCA1 and ABCG1 in the overall process of cellular cholesterol efflux.
