    • ••] loci. It was shown that V and (D)/J gene segments are brought into close proximity by a DNA looping mechanism. Furthermore, Roldan et al. also report that IgH locus contraction and centromeric recruitment are reversed after pre-B cell receptor signaling, suggesting a novel mechanism to mediate allelic exclusion.

    • H.E. Liang, L.Y. Hsu, D. Cado, and M.S. Schlissel Variegated transcriptional activation of the Immunoglobulin κ locus in pre-B cells contributes to the allellic exclusion of light-chain expression Cell 118 2004 19 29 Knockin mice were generated in which the GFP cDNA was placed under control of the Igκ germline promoter. The authors reports that only a small percentage of pro-B cells express the GFP marker, that this expression is monoallelic and that the transcribing Jκ alleles are the preferred targets for Vκ-to-Jκ recombination. This study supports a model in which a stochastic mechanism leads to monoallelic expression of the IgJκ locus, thus contributing to the establishment of allelic exclusion.

    • Regulation of T cell receptor beta allelic exclusion at a level beyond accessibility
    • A. Jackson, H.D. Kondilis, B. Khor, B.P. Sleckman, and M.S. Krangel Regulation of T cell receptor beta allelic exclusion at a level beyond accessibility Nat Immunol 6 2005 189 197 In this knock-in study, the TCRα gene enhancer was introduced downstream of the Vβ12 gene segment. Despite increased Vβ accessibility and germline transcription in double-positive thymocytes, the inhibition of Vβ-to-DJβ rearrangement appears to be maintained. Following flow cytometry analysis to identify double TCRβ-expressing cells, the authors conclude that allelic exclusion at Vβ12 was partially subverted in double-negative thymocytes.
