The data show a 16-nm periodicity, indicating that kinesin-1 spends most of its time with its heads 16 nm apart, or alternatively indicating that the labelled molecule is limping.

The rate of back-stepping is dependent on ATP concentration, but is insensitive to the size of the backwards load. Step averaging is used to show that both forward and back steps are single events that occur on the microsecond time scale. The duration of these events is dependent on bead size, indicating that the measured step time is limited by bead diffusion.
