    • Targeting Schwann cells by nonlytic arenaviral infection selectively inhibits myelination
    • Selective targeting of α-dystroglycan on Schwann cells by LCMV and Lassa fever virus from the non-lytic Arenavirus family was demonstrated. Persistent LCMV infection rendered immature Schwann cells incapable of forming mature myelin sheath during differentiation, and caused demyelination in mature Schwann cells that form myelin sheaths. A correlation between the disorganization of the laminin-2 network and downregulation of α-dystroglycan with myelin defects was demonstrated. Viral manipulation of dystroglycan and laminin-associated functions in Schwann cells was proposed for myelination and demyelination. The study also shows the use of LCMV as a tool to dissect the molecular basis of myelination.

Slowed nerve conduction and nodal changes, including reduced sodium channel density and disorganized microvilli, were observed in mutant mice. Data suggest that dystroglycan is crucial for both myelination and normal maintenance of voltage-gated sodium channels at nodes of Ranvier, possibly by mediating trans interactions between Schwann cell microvilli and the nodal axolemma.
