A similar prolongation of NMDA receptor EPSCs is observed in paired CA3-CA1 pyramidal cell recordings during GluT inhibition, although the effect was only apparent during trains of stimuli. These results indicate that GluTs restrict activation of extrasynaptic receptors and prevent spillover at Schaffer collateral synapses in the hippocampus.

    • This study investigated the source of glutamate that activates AMPA receptors in Bergmann glia as a result of climbing fiber stimulation. The authors were able to resolve quantal EPSCs in Bergmann glial cells, and found that these events were not coincident with quantal EPSCs recorded simultaneously from Purkinje cells. These results indicate that the glutamate that reaches Bergmann glial AMPA receptors arises from distinct release events, rather than spillout from climbing fiber-Purkinje cell synapses. These data suggest that Bergmann glial AMPA receptors are activated by vesicle fusion at ectopic sites, where the climbing fiber terminal and the glial membrane are in direct apposition.

    • Spillover of glutamate onto synaptic AMPA receptors enhances fast transmission at a cerebellar synapse
    • Synapses within the mossy fiber glomerulus are not separated by glial processes. The authors show that as much as half of the charge transfer associated with evoked AMPA EPSCs is produced by spillover of glutamate from neighboring synapses within the glomerulus. This spread of glutamate serves to increase synaptic efficacy and improve the reliability of transmission at these synapses.
