Fibroblasts are transfected with YFP-actin and GFP-actin and the YFP signal bleached in a defined area. The difference and the ratio of the signals provide a read-out of the redistribution of monomeric actin in the bleached region. Measurements show that actin as far as 12 μm behind the lamellipodium is transported to the lamellipodium front at a rate faster than can be explained by diffusion. The authors propose that myosin II sets up a pressure gradient to drive the flow.

The maintenance of the shape of the lateral wings is shown to arise from the combination of decreased polymerization and increased retrograde flow. It is also demonstrated that retrograde flow can be readily visualized by enhanced phase contrast microscopy.
