Silencing of the mitogen-activated protein kinase MPK6 compromises disease resistance in Arabidopsis
-
F.L. Menke, J.A. van Pelt, C.M. Pieterse, and D.F. Klessig Silencing of the mitogen-activated protein kinase MPK6 compromises disease resistance in Arabidopsis Plant Cell 16 2004 897 907 The authors present data showing that AtMPK6 plays a role in R-gene-mediated and basal defense. Significantly, the AtMPK6-silenced lines were not affected in their ability to develop SAR or ISR. This work sheds more light on the role of AtMPK6 in resistance, and provides insight into the regulation of defense pathways downstream of MAPKs.

MAPKKKα is a positive regulator of cell death associated with both plant immunity and disease
-
O. del Pozo, K.F. Pedley, and G.B. Martin MAPKKKα is a positive regulator of cell death associated with both plant immunity and disease EMBO J 23 2004 3072 3082 Using VIGS, LeMAPKKKα, a MAPKKK from tomato was found to play a role in cell death in both the resistance response and during infection by virulent pathogens. Epistasis experiments placed LeMAPKKKα upstream of the tomato homologs of NtMEK2 and NtSIPK. The authors propose that LeMAPKKKα acts as a key molecular switch that regulates signal transduction pathways that are associated with plant immunity and disease susceptibility.

K.F. Pedley, and G.B. Martin Identification of MAPKs and their possible MAPKK activators involved in the Pto-mediated defense response of tomato J Biol Chem 279 2004 49229 49235 This paper provides key biochemical data that identify the MAPKs that are elicited as part of the Pto-mediated defense response. This paper complements other studies that use VIGS to place MAPKs downstream of Pto. The authors also identify a MAPKK from a previously uncharacterized MAPKK family that appears to play a positive role in cell death.

Phosphorylation of 1-aminocyclopropane-1-carboxylic acid synthase by MPK6, a stress-responsive mitogen-activated protein kinase, induces ethylene biosynthesis in Arabidopsis
-
Y. Liu, and S. Zhang Phosphorylation of 1-aminocyclopropane-1-carboxylic acid synthase by MPK6, a stress-responsive mitogen-activated protein kinase, induces ethylene biosynthesis in Arabidopsis Plant Cell 16 2004 3386 3399 In an elegant study using both gain- and loss-of-function analysis, the authors provide both biochemical and genetic data that describe at least one role played by AtMPK6 in plant defense. AtMPK6 was shown to phosphorylate two isoforms of ACS directly. The phosphorylation leads to the stabilization of ACS, a short-lived enzyme, and results in an increase in ethylene biosynthesis. This is the first paper to provide in vivo data for the phosphorylation of a plant MAPK substrate, and sheds light on both plant defense and stress-related ethylene biosynthesis.
