The induction of SAR resulted in the reduction of NPR1 to an active monomeric form that localized in the nucleus. Mutational analyses indicated that these properties are regulated by the reduction of two conserved cysteines in NPR1. The authors also show that treatment with the SA analog INA results in an overall reduction in the cellular redox potential, as measured by the levels and ratio of reduced/oxidized glutathione.

Fobert The Arabidopsis NPR1 disease resistance protein is a novel cofactor that confers redox regulation of DNA binding activity to the basic domain/leucine zipper transcription factor TGA1 Plant Cell 15 2003 2181 2191 Using an in planta transient expression assay that is conceptually similar to the yeast two-hybrid system, the authors show that TGA1 interacts with NPR1 in Arabidopsis leaves upon SA treatment. Domain swapping and site-directed mutagenesis identified two conserved cysteines in TGA1 and TGA4 that, when oxidized, interfere with NPR1 interaction. Monitoring the redox status of TGA1/TGA4 cysteines in vivo revealed that SA induced their reduction. In vitro studies indicated that the DNA binding activity of TGA1 is regulated only indirectly by redox conditions through the NPR1 cofactor.

Transient overexpression of CITRX compromised Cf-9/Avr9-dependent defense responses. The authors propose a model by which CITRX could negatively regulate Cf-9 function.
