This work highlights the molecular basis of at least one mechanism for chloroplast-to-nucleus signalling. The authors show that the tetrapyrrole intermediate Mg-protoporphyrin (Mg-ProtoIX) acts as a molecule that signals between the chloroplast and nucleus. Accumulation of Mg-ProtoIX is both necessary and sufficient to regulate the expression of many nuclear genes encoding chloroplastic proteins that are associated with photosynthesis.

Excellent work in which the authors illustrate the role of red-light-sensing mechanisms in plant pathogen interactions. The induction of PR1 by SA and its functional analogues was found to correlate strictly with the activity of the signaling pathway controlled by the PHYA and PHYB photoreceptors. These findings presumably represent only the tip of the iceberg of the many-fold influences of light-dependent signal transduction on plant disease defence mechanisms.

The authors present important findings which demonstrate that the SA synthesis pathway, or part of this pathway, is localised in the chloroplast. Figure 2 of this review clearly shows that SA levels increase during acclimation to high light and suggests that SA plays a role in acclimation to EEE. Therefore, SA does not appear to be a specific signalling molecule for disease defence responses alone. The full spectrum of SA functions and the nature of SA signalling in plants are not yet clear and remain to be elucidated.
