Three cdg operons control cellular turnover of cyclic di-GMP in Acetobacter xylinum: genetic organization and occurrence of conserved domains in isoenzymes. J. Bacteriol. 180:1998;4416-4425 In this work, the authors used reverse genetics to identify three operons that are involved in controlling the cellular levels of c-diGMP in G. xylinum. Each operon contains two orthologs, coding for a PDE and a DGC, respectively. Sequence analysis revealed that all gene products contain highly conserved and bacteria-specific GGDEF and EAL domains.

Lett. 203:2001;11-21 In this 'mini review', the authors perform an in silico analysis of bacterial genomes, with respect to two-component signal transduction systems. The discovery of a compound domain architecture of sensor kinases and response regulators with several novel conserved domains, such as PAS, GAF [cGMP-specific and -stimulated phosphodiesterases/ adenylate cyclases (Anabaena)/ FhlA (E. coli)], HAMP, GGDEF, EAL, and HD-GYP begins to shed light on a novel complexity of signal transduction pathways in bacteria.

PleD is shown to act downstream of several components (PleC, DivJ, DivK) that constitute a two-component signal transduction network that is crucial for Caulobacter asymmetric cell division and differentiation.

Genes Dev 2004, in press. This paper explicitly demonstrates that the GGDEF domain of PleD is a DGC, which is activated upon phosphorylation of the receiver domain. Moreover, evidence is presented to suggest that the production of c-diGMP is spatially restricted in the cell. The PleD response regulator dynamically localizes to the cell pole, where the morphogenetic changes take place. Targeting of PleD to the pole is dependent on its phosphorylation and on the presence of the cognate kinases PleC and DivJ.

FEMS Microbiol. Lett. 204:2001;163-167 This paper shows that cellulose production in R. leguminosarum and A. tumefaciens is induced upon expression of several genes from different bacterial species, all of which code for GGDEF proteins. Because the GGDEF domain is the only part that is common to all of the gene products investigated, the authors conclude that this domain is responsible for c-diGMP synthesis.
