In this way, the composition of the top of the layer can be determined. In this paper, researchers used this to reliably measure whether a protein has denatured upon adsorption to the surface. Key to this measurement is the preservation of the protein conformation with trehalose before exposure to ultrahigh vacuum conditions for the measurement.

This study is important because it introduces a method to control the specific chemical nature of the solid surface. In addition, it shows that, in the cases studied, the degree of unfolding depends on the concentration of hydrophobic groups on the surface and on the concentration of protein in solution.
