    • Requirement of signalling by receptor tyrosine kinase RET for the directed migration of enteric nervous system progenitor cells during mammalian embryogenesis
    • This study establishes a correlation between anteroposterior dynamics of the expression of ret by migrating NCCs and GDNF (ret ligand) in the gut mesenchyme. The results are consistent with a chemotactic role for this growth factor signaling in ensuring the progressive colonization of the intestine by enteric NC precursors.

    • Neural crest stem cells undergo cell-intrinsic developmental changes in sensitivity to instructive differentiation signals
    • This study provided evidence that specification of autonomic neurons into noradrenergic/cholinergic subtypes is controlled, at least in vitro, by differential responses of NCSCs to BMP2 concentrations. In addition, the sensitivity of rat neural tube- and sciatic nerve-derived NCSCs to external cues was analyzed both in vivo and in vitro. It was found that nerve NCSCs are biased toward a gliogenic fate and can differentiate only into parasympathetic-subtype neurons after transplantation into the NC migration pathway of E2.5 chick. The decreased ability of nerve NCSCs to yield sympathetic neurons was related to a lower BMP2 sensitivity.

    • Molecular control of ciliary neuron development: BMPs and downstream transcriptional control in the parasympathetic lineage
    • The authors show that the development of parasympathetic neurons of the chick ciliary ganglion is controlled by BMPs produced in the NCC environment, as in the case of sympathetic neurons. Unlike sympathetic neuron precursors, however, those of the ciliary ganglion express only transiently noradrenergic markers and adopt a cholinergic phenotype. Cholinergic versus noradrenergic differentiation is related to expression of dHand transcription factor only in sympathetic neurons, suggesting that dHand maintains activation of noradrenergic genes in autonomic progenitors.

    • Using an inducible Cre-recombinase fate-mapping system, it is shown that ngn2-expressing cells in the early NC are strongly, although incompletely, biased to a sensory fate. However, in the developing DRG, these cells have the same potential to differentiate as either glia or neurons than the bulk of the NCC population, meaning that segregation of glial and neuronal progenitors occurs after initial specification to the sensory lineage.

    • Functional studies were performed using dissociated cultures of rat neural tubes where NCCs are able to yield both autonomic and sensory neurons and can be identified by the Sox10 NC marker. Overexpressing ngn2 and Mash1 could drive these early NCCs to sensory and autonomic fates, respectively. However, misexpression of ngn2 was no longer able to promote a sensory identity in cultured migratory NCSCs or in early NCCs submitted to high concentrations of BMP2 and, hence, these cells acquired an autonomic fate. Therefore, Mash1 and ngn2 display differential sensitivity to growth factor and cell developmental state.

    • Gain-of-function experiments on cultured NCSCs were devised to analyze feedback signals mediated by Notch/Delta and modulated by intracellular Numb. Misexpression of Notch and Numb in NCSCs had opposite effects on gliogenic response to soluble Delta. NCSC responsiveness to Delta spontaneously increased in the clones as a result of cell-cell interactions and can be abrogated if the clonal progeny is maintained at low cell density. In addition, this study revealed that nerve NCSCs display increased responsiveness to Delta as compared to early NCSCs, which mediates their bias toward gliogenesis.

    • Reversal of developmental restrictions in neural crest lineages: Transition from Schwann cells to glial-melanocytic precursors in vitro
    • In this study, we challenged the stability of the glial cell phenotype by exposure to ET3 in vitro. Schwann cells were immuno-selected from E10.5 quail sciatic nerves and grown in clonal cultures. These cells gave rise to a mixed progeny of glial cells and pigment cells and/or melanoblasts in response to ET3 mitogenic signal. Therefore, differentiated glial cells and melanocytes [90] in vitro can reverse to their common bipotent ancestor upstream in NC lineage hierarchy.

    • SOX10 maintains multipotency and inhibits neuronal differentiation of neural crest stem cells
    • In addition to its known function in peripheral gliogenesis, the HMG transcription factor SOX10 was shown here to influence NCSC properties in several ways: constitutively expressed SOX10 maintained neurogenic and gliogenic potentials, inhibited differentiation and prevented proliferation arrest of rat NCSC in vitro, suggesting that this protein is an important regulator of NCC multipotency.
