K114 is an analog of Congo red, and interactions of K114 with amyloid fibrils results in quantitative increases in fluorescence (32). Five μl of each sample was added to 100 μl of 50-μM K114 in 100 mM glycine, pH 8.5, and fluorescence (excitation wavelength 380 nm, emission wavelength = 550 nm, cutoff = 530 nm) was measured with a SpectraMax Gemini fluorometer and SoftMax Pro 4.0 software.
