This interesting study demonstrated a reduced frequency of the TaqlB B2 allele in patients with established coronary heart disease and HDL deficiency suggesting that increased HDL cholesterol levels resulting from decreased CETP activity are associated with lower coronary heart disease risk. Patients with two B2 alleles had the least favorable plasma lipid profile but showed the greatest lipid lowering response to gemfibrozil.

This is the first and only single nucleotide polymorphism haplotype study to date concerning six relevant lipid metabolism genes and their association with lipid levels. A clear association was found between CETP haplotypes and HDL cholesterol and HDL/LDL ratio promising a new tool for identifying the genetic background for atherosclerosis.

This study provides evidence for the presence of an unpaired cysteine residue at the active site of cynomolgous monkey CETP. Conclusively, CETP inhibitors targeting this residue may act by producing steric hindrances for cholesteryl ester transfer activity.

This interesting study not only described a three-fold increased plasma concentration of lipid transfer inhibitor protein in hypercholesterolemic patients, but also a negative correlation between levels of LTIP and triglyceride in plasma possibly resulting from redistribution of LTIP from LDL to very low-density lipoprotein and subsequent increased clearance from the circulation.

Treatment of healthy subjects with CETP inhibitor JTT-705 led to the desired changes in lipoprotein profile including increase of HDL cholesterol and decrease of LDL cholesterol after 4 weeks.
