SNP judgments and freedom of association
-
Hegele RA. SNP judgments and freedom of association [editorial]. Arterioscler Thromb Vasc Biol 2002; 22:1058-1061. This invited editorial brings to our attention many of the issues that we have to consider when interpreting the literature for association studies. Moreover, it points out the proper experimental approaches in order to avoid many of the common pitfalls in this type of study.

Positive results in association studies are associated with departure from Hardy-Weinberg equilibrium: Hint for genotyping error?

Xu J, Turner A, Little J, et al. Positive results in association studies are associated with departure from Hardy-Weinberg equilibrium: hint for genotyping error? Hum Genet DOI 10.1007/s00439-002-0819-y. We all know about Hardy-Weinberg equilibrium, but sometimes we forget that it exists. This paper describes the many instances in which Hardy-Weinberg equilibrium is not present in case control studies describing significant associations. This is a short paper that should be read by all of us doing this type of study.

Dietary fat intake determines the effect of a common polymorphism in the hepatic lipase gene promoter on HDL metabolism: Evidence of a strong dose-effect in this gene-nutrient interaction in the Framingham Study
-
Ordovas JM, Corella D, Demissie S, et al. Dietary fat intake determines the effect of a common polymorphism in the hepatic lipase gene promoter on HDL metabolism: evidence of a strong dose-effect in this gene-nutrient interaction in the Framingham Study. Circulation 2002; 106:2315-2321. This research demonstrates a strong interaction between this LIPC polymorphism and dietary fat. This could be one of the reasons why some ethnic groups have problems adapting to more westernized dietary habits.

Frequent mutation in the ABCC6 gene (R1141X) is associated with a strong increase in the prevalence of coronary artery disease

A very interesting observation that further boosts the interest in the ABC family of genes in relation to CVD. Very strong results, but it needs to be replicated in other populations. Moreover, the allele frequency of this polymorphism is low and it may not have a strong effect on the total population risk.

Heterogeneity of linkage disequilibrium in human genes has implications for association studies of common diseases
-
Tiret L, Poirier O, Nicaud V, et al. Heterogeneity of linkage disequilibrium in human genes has implications for association studies of common diseases. Hum Mol Genet 2002; 11:419-429. We need better definition of the alleles that are associated with relevant phenotypes. One way to achieve this is by examining multiple markers per loci and to derive haplotypes. The success of this approach depends on the extent of linkage disequilibrium in the region being considered. These authors have examined this issue in great detail for several CVD candidate genes.
