RXR and LXR agonists were found to increase expression of the ABC transporters A1 and G1 in Caco-2 cells, a human intestinal cell line. Moreover, treatment with RXR and LXR agonists enhanced apolipoprotein-mediated cholesterol efflux from the basolateral, but not apical, membranes of these cells.

This is the first study to report the effects of ABCA1 overexpression in mice. Overexpression of ABCA1 in the liver caused a significant increase in biliary cholesterol and phospholipid content, indicating that ABCA1 may facilitate the transport of these lipids into the bile.

In this study, treatment with an LXR agonist significantly increased the secretion of cholesterol, but not phospholipid, into the bile, of both ABCA1 knockout and wild type mice. Taken together with the results of their earlier work [26•], this report provides further evidence that the mechanism(s) responsible for the LXR-induced hypersecretion of cholesterol into the bile may be independent of ABCA1.
