    • Genetic control of acute ethanol-induced behaviors in Drosophila
    • This paper describes the effects of ethanol on the speed and direction of fly walking behavior using simple line-crossing assays. These parameters of ethanol-induced behavior are altered by two ethyl methane sulfonate-induced mutants (isolated originally through their abnormal behavior in the inebriometer), which shows that the effects of ethanol on postural control and on walking behavior are mechanistically related.

    • Functional ethanol tolerance in Drosophila
    • This paper is the first description of the phenomenon of functional ethanol tolerance in Drosophila. It provides a detailed analysis of the kinetics of tolerance development and dissipation, and the underlying dose-response relationships. This study also defines a small group of neurons of the central complex that are necessary for normal development of tolerance. In addition, a mutant strain lacking octopamine shows strongly reduced tolerance, indicating that this neuromodulator is required in the process.

    • This study shows that feeding flies the tyrosine hydroxylase inhibitor 3IY, which causes a 90% reduction in dopamine levels as measured by high-performance liquid chromatography, causes flies to become resistant to cocaine, nicotine and to the stimulating (but not sedating) effects of ethanol. For cocaine, which is known to inhibit dopamine reuptake into presynaptic terminals, this result is perhaps not surprising. How dopamine modulates the behavioral effects of nicotine and ethanol is not obvious; these drugs may act directly on dopamine cells or elsewhere in the neural circuits that regulate these behaviors.

    • PKA-RII-deficient Drosophila are viable but show developmental, circadian and drug response phenotypes
    • This study describes the molecular, biochemical and behavioral characterization of a Drosophila strain expressing extremely low levels of PKA-RII. In these flies, the low basal PKA activity is increased two-fold but cAMP-stimulated PKA activity is strongly reduced. The mutant flies show decreased sensitivity to the sedative effects of ethanol, a failure to sensitize to cocaine, and abnormal circadian rhythms.

The quinpirole-induced locomotor response of these headless flies changes as a function of time of day in light/dark conditions; however, this modulation is substantially reduced in constant light conditions and completely absent in period mutant flies under constant light conditions. In addition, this circadian modulation of dopamine responsiveness is still seen if flies are tested 12 hours after decapitation, showing that the head pacemaker is, at least in part, dispensable for this modulation. The authors suggest that quinpirole-sensitive amine receptors are involved in the motor output pathway of the circadian clock.
