Boll M., Fuchs G., Meier C., Trautwein A.X., Lowe D.J. EPR and Mössbauer studies of benzoyl-CoA reductase. J Biol Chem. 275:2000;31857-31868. Using EPR and Mössbauer spectroscopy, three [4Fe-4S] clusters of benzoyl-CoA reductase are identified as cofactors. The unique reversible switch from the S = 1/2 to the S = 7/2 state during ATP-hydrolysis is demonstrated by Mössbauer spectroscopy.

Crystal structure of the Acidaminococcus fermentans 2-hydroxyglutaryl-CoA dehydratase component A

The crystal structure of the ATP-driven electron activating 'activase' from A. fermentans provides important structural information for benzoyl-CoA reductase as both enzymes show high amino acid similarities. In spite of non-existing amino acid similarities, the structured greatly resembles nitrogenase iron protein. Two 'cluster helices' are identified that are supposed to be involved in major conformational changes during ATP hydrolysis.

-
Unciuleac M., Boll M. Mechanism of ATP-driven electron transfer catalyzed by the benzene ring-reducing enzyme benzoyl-CoA reductase. Proc Natl Acad Sci USA. 98:2001;13619-13624. Möbitz H., Boll M. A Birch-like mechanism in enzymatic benzoyl-CoA reduction - a kinetic study of substrate analogues combined with an ab initio model. Biochemistry. 41:2002;1752-1758. In this study, evidence is provided that benzoyl-CoA reductase catalysis proceeds in a Birch-like mechanism. A partial protonation of the carbonyl oxygen and a simultaneous electron/proton transfer are considered to facilitate reduction of the aromatic ring.

-
Rabus R., Wilkes H., Behrends A., Armstroff A., Fischer T., Pierik A.J., Widdel F. Anaerobic initial reaction of n-alkanes in a denitrifying bacterium: evidence for (1-methylpentyl)succinate as initial product and for involvement of an organic radical in n-hexane metabolism. J Bacteriol. 183:2001;1707-1715. The fumarate addition product of n-alkanes is identified and the involvement of a glycyl-radical enzyme in the process is shown by EPR-spectroscopy.
