These authors identified ABCA1 as the gene for Tangier disease using expression microarray analysis and presented several lines of evidence that this protein controls the apolipoprotein-mediated lipid removal pathway. They also showed that ABCA1 expression was regulated by both cholesterol and cAMP in fibroblasts, and that this protein appears on the cell surface.

Nat Genet 2000; 24:192-196. This is the first description of the phenotype of ABCA1 knockout mice, which closely resembles that of patients with Tangier disease. These mice will be useful in defining the role of ABCA1 in lipoprotein metabolism and atherogenesis. This study also confirms that ABCA1 is localized to the cell surface as well as the Golgi and presents evidence that mutations in ABCA1 impair lipid transport from the Golgi to the plasma membrane.

[39] showing that cAMP analogs markedly induce apolipoprotein-mediated lipid removal from several murine macrophage lines but not from many other cultured cell lines. It also shows that apolipoprotein A-I directly binds to a 200 kD cell-surface protein that is induced by cAMP, providing the first hint that apolipoprotein A-I might bind directly to ABCA1.

This comprehensive study uses different apolipoproteins, synthetic peptides, and deletion mutants to show that insertion of amphipathic helices into phospholipid bilayers is a required step for lipid removal by apolipoproteins. These findings also implicate lipid affinity as a critical determinant for apolipoprotein specificity.
