Targeted mutation reveals a central role for SR-BI in hepatic selective uptake of high density lipoprotein cholesterol

Targeted mutation reveals a central role for SR-BI in hepatic selective uptake of high density lipoprotein cholesterol. Proc Natl Acad Sci U S A 1998; 95:4619-4624. A mouse model with attenuated SR-BI expression showed increased HDL cholesterol levels and decreased clearance of HDL cholesterol through the selective lipid uptake pathway, confirming previous results derived from a SR-BI null mouse [42].

Hepatic scavenger receptor BI promotes rapid clearance of high density lipoprotein free cholesterol and its transport into bile

Hepatic scavenger receptor BI promotes rapid clearance of high density lipoprotein free cholesterol and its transport into bile. J Biol Chem 1999; 274:33398-33402. Regulation of adrenal scavenger receptor-BI expression by ACTH and cellular cholesterol pools
-
50 Sun Y, Wang N, Tall AR. Regulation of adrenal scavenger receptor-BI expression by ACTH and cellular cholesterol pools. J Lipid Res 1999; 40:1799-1805. This report demonstrates that SR-BI expression is regulated by cholesterol and ACTH in vitro and in vivo. Moreover, cholesterol depletion can induce adrenal SR-BI expression independently of plasma ACTH levels, although in vivo this effect is apparently outweighed by ACTH.

This study strongly supports a role for SF-1 in regulating SR-BI expression during the development of steroidogenic tissues. Furthermore, this work shows that SR-BI expression in cultured adrenocortical cells is regulated by cholesterol and trophic hormones, although cholesterol-dependent regulation can be overridden by trophic factors.

Expression and microvillar localization of scavenger receptor, class B, type I (a high density lipoprotein receptor) in luteinized and hormone-desensitized rat ovarian models. Endocrinology 1998; 139:2847-2856. These studies show that SR-BI can be regulated by gonadotrophin and cholesterol status in luteinized granulosa cells. In addition, at least some of the SR-BI expression was found in the microvilli and microvillar channels of the luteal cell surface, which are specialized regions for selective lipoprotein cholesterol uptake in steroidogenic cells.

The class B, type I scavenger receptor promotes the selective uptake of high density lipoprotein cholesterol ethers into caveolae
-
85 Graf GA, Connell PM, van der Westhuyzen DR, Smart EJ. The class B, type I scavenger receptor promotes the selective uptake of high density lipoprotein cholesterol ethers into caveolae. J Biol Chem 1999; 274.12043-12048. Using radiolabelled cholesterol ether as a tracer of HDL lipids, this study concludes that plasma membrane caveolae are acceptors for HDL-derived cholesteryl esters and that caveolae constitute a reversible plasma membrane pool from which cholesterol is internalized to intracellular compartments.

Comparison of class B scavenger receptors, CD36 and scavenger receptor BI (SR-BI), shows that both receptors mediate high density lipoprotein-cholesteryl ester selective uptake but SR-BI exhibits a unique enhancement of cholesteryl ester uptake
-
88 Connelly MA, Klein SM, Azhar S, Abumrad NA, Williams DL. Comparison of class B scavenger receptors, CD36 and scavenger receptor BI (SR-BI), shows that both receptors mediate high density lipoprotein-cholesteryl ester selective uptake but SR-BI exhibits a unique enhancement of cholesteryl ester uptake. J Biol Chem 1999; 274:41-47. Results of these studies show that SR-BI-mediated high affinity HDL binding to the cell surface is not sufficient to ensure efficient cellular selective lipid uptake from HDL. The extracellular domain of SR-BI is essential for HDL cholesteryl ester uptake through the selective uptake process.

Mechanism of scavenger receptor class B type I-mediated selective uptake of cholesteryl esters from high density lipoprotein to adrenal cells
-
89 Rodrigueza WV, Thuahnai ST, Temel RE, Lund-Katz S, Phillips MC, Williams DL. Mechanism of scavenger receptor class B type I-mediated selective uptake of cholesteryl esters from high density lipoprotein to adrenal cells. J Biol Chem 1999; 274:20344-20350. This study shows a low activation energy for selective cholesteryl ester uptake from native or reconstituted HDL to cells, indicating that this process occurs via a non-aqueous pathway. The authors propose that SR-BI may facilitate selective cholesterol uptake through the formation of a hydrophobic channel through which cholesteryl ester molecules move down a chemical gradient from HDL particles to the plasma membrane.

Hepatic lipase promotes the selective uptake of high density lipoprotein-cholesteryl esters via the scavenger receptor B1. J Lipid Res 1999; 40:1294-1303. Remodeling of HDL by CETP in vivo and by CETP and hepatic lipase in vitro results in enhanced uptake of HDL CE by cells expressing scavenger receptor B-I. J Lipid Res 1999; 40:1185-1193. This study reports that the remodeling of HDL by CETP followed by the action of hepatic lipase increased uptake of HDL cholesterol by SR-BI, suggesting that the selective uptake and CETP pathways may act synergistically in HDL metabolism in vivo.
