Hepatic lipase mediates the uptake of chylomicrons and β-VLDL into cells via the LDL receptor-related protein (LRP)
-
Krapp A, Ahle S, Kersting S, Hua Y, Kneser K, Nielsen M, et al. Hepatic lipase mediates the uptake of chylomicrons and β-VLDL into cells via the LDL receptor-related protein (LRP). J Lipid Res 1996; 37:926-936. This manuscript provides evidence supporting a role of HL in cellular chylomicron and β-VLDL uptake via LRP. Crosslinking experiments confirm binding of HL to LRP and competition studies indicate that lipoproteins most likely interact with the carboxyl-terminal domain of HL.

Heparan sulfate proteoglycans participate in hepatic lipase and apolipoprotein E-mediated binding and uptake of plasma lipoproteins, including high density lipoproteins
-
Ji Z-S, Dichek HL, Miranda RD, Mahley RW. Heparan sulfate proteoglycans participate in hepatic lipase and apolipoprotein E-mediated binding and uptake of plasma lipoproteins, including high density lipoproteins. J Biol Chem 1997; 272:31285-31292. The role of proteoglycans and RAP sensitive pathway(s) as well as catalytically inactive hepatic lipase in mediating the uptake of β-VLDL, IDL and HDL is evaluated in transfected rat hepatoma cells. Evidence that hepatic lipase enhances both direct uptake of HDL particles and selective uptake of HDL cholesteryl esters via different pathways is provided.

Identification of scavenger receptor SR-BI as a high density lipoprotein receptor
-
Acton S, Rigotti A, Landschulz KT, Xu S, Hobbs HH, Krieger M. Identification of scavenger receptor SR-BI as a high density lipoprotein receptor. Science 1996; 271:518-520. This paper represents a major breakthrough in the HDL receptor field. The authors identify and characterize the murine scavenger receptor, SR-BI, which mediates the selective uptake of cholesteryl esters from HDL.

Scavenger receptor BI (SR-BI) is up-regulated in adrenal gland in apolipoprotein A-I and hepatic lipase knock-out mice as a response to depletion of cholesterol stores
-
Wang N, Weng W, Breslow JL, Tall AR. Scavenger receptor BI (SR-BI) is up-regulated in adrenal gland in apolipoprotein A-I and hepatic lipase knock-out mice as a response to depletion of cholesterol stores. J Biol Chem 1996; 271:21001-21004. Evidence that SR-BI expression is regulated by intracellular cholesterol is provided by analysis of adrenal SR-BI mRNA in apoA-I and hepatic lipase deficient mice with decreased cholesterol adrenal stores.

Recent study examining coronary heart disease and all cause mortality of 8000 Israeli men. Hepatic lipase deficiency increases plasma cholesterol but reduces susceptibility to atherosclerosis in apolipoprotein E-deficient mice
-
Mezdour H, Jones R, Dengremont C, Castro G, Maeda N. Hepatic lipase deficiency increases plasma cholesterol but reduces susceptibility to atherosclerosis in apolipoprotein E-deficient mice. J Biol Chem 1997; 272:13570-13575. Hepatic lipase deficiency in female apoE deficient mice leads to significant increases in plasma cholesterol, β-VLDL and HDL concentrations as well as reductions in aortic atherosclerosis. HDL from apoE and hepatic lipase deficient mice had increased capacity to promote cholesterol efflux. These findings suggest that hepatic lipase may modulate the development of atherosclerosis by alternative pathways independent of changes in the plasma lipid profile.
