A detailed molecular belt model for apolipoprotein A-I in discoidal high density lipoprotein
-
16 Segrest JP, Jones MK, Klon AE, Sheldahl CJ, Hellinger M, De Loof H, et al. A detailed molecular belt model for apolipoprotein A-I in discoidal high density lipoprotein. J Biol Chem 1999; 274:31755-31758. A detailed model for the lipid-associated carboxyl-terminal domain of apolipoprotein A-I is constructed based upon the assumption that lipid imposes profound constraints on the conformation and orientation of proteins associated with it. In the model, two apolipoprotein A-I molecules are wrapped belt-wise around a small discoidal patch of bilayer containing 160 lipid molecules. A striking 11mer/22mer periodicity in perfectly aligned interhelical salt bridges was found. The structure with the most favorable alignment of salt bridges exactly matches that of the (nonplanar) crystal structure of lipid-free apolipoprotein A-I.

Mutations in ABC1 were detected in both Tangier disease and familial HDL deficiency, indicating that the two are allelic. The authors suggest that the protein encoded by ABC1 is a key element in influencing intracellular cholesterol transport.

Mutations in the ABC1 gene in familial HDL deficiency with defective cholesterol efflux

Three French-Canadian families and one Dutch family with familial HDL deficiency were studied. A different mutation was detected in ABC1 in each family. Each mutation either created a stop codon predicted to result in truncation of ABC1, or altered a conserved amino acid residue.

Biochemistry 1999; 38:1727-1735. In-vitro experiments showed gradual association of phospholipid with apolipoprotein A-I leading to the formation of an unstable lipoprotein A-I particle containing one apolipoprotein A-I molecule, which is an efficient substrate for LCAT. The interaction of lipoprotein A-I with LCAT suggests direct apolipoprotein A-I-LCAT interactions.

Expression of serum amyloid A protein in the absence of the acute phase response does not reduce HDL cholesterol or apoA-I levels in human apoA-I transgenic mice. J Lipid Res 1999; 40:648-653. This recent study showed that high plasma levels of SAA in the absence of a generalized acute-phase response does not result in reduction of HDL-cholesterol or apolipoprotein A-I.
