The results support the idea that internalized V2 receptors are stably retained in endocytic vesicles and that this trafficking fate causes delayed resensitization of signal transduction, thereby suggesting a novel function of beta-arrestins in GPCR regulation

    • Biochemical and immunocytochemical assays were used to examine the effects of mutations of the cytoplasmic tail of the B2AR on endocytic trafficking. Specific protein interactions involving the cytoplasmic tail of the receptor and the cortical actin cytoskeleton were observed to promote rapid recycling of the B2AR. The results suggest a novel role of PDZ domain-mediated interactions with NHERF family protein(s) in endocytic sorting of GPCRs; they also suggest a role of receptor phosphorylation in regulating this mechanism
