    • Role of Niemann-Pick type C1 protein in intracellular trafficking of low density lipoprotein-derived cholesterol
    • In contrast to existing views, this study suggests that NPC1 is involved in regulating recycling of plasma-membrane-derived cholesterol, rather than egression of cholesterol, following hydrolysis of LDL-mediated cholesterol delivery. The latter reaches the cell surface, and it is the subsequently reinternalized cholesterol fraction that depends on NPC1 for recycling. Accordingly, NPC1 may discriminate between different cholesterol pools.

    • Exploiting the partitioning of lipid probes into membrane domains of distinct fluidity, they were used to define the molecular parameters (which are correlated to membrane biophysical properties) that determine the sorting of lipids and lipid-attached compounds in eukaryotic cells. The overall findings are consistent with the effect of membrane rigidity (also accomplished by modulating the cellular pool of cholesterol) on regulating recycling versus endosomal/lysosomal trafficking pathways.

    • Maturation of the axonal plasma membrane requires upregulation of sphingomyelin synthesis and information of protein-lipid complexes
    • A fascinating study that shows that the level of sphingomyelin synthesis correlates with 'raft' formation and the subsequent sorting and targeting of a GPI-anchored protein and a ganglioside. The latter two are targeted to the ('apical') axonal plasma membrane during a late developmental stage during neuronal maturation. It can be inferred that sphingomyelin and cholesterol induce raft biogenesis in this case, whereas a role for glycosphingolipids in raft assembly and stability is rather unlikely.
