24. C. Bordier, J. Biol. Chem. 256, 1604 (1981). * Precondensed Triton X-114 is obtained by subjecting a 2% (v/v) solution of commercial-grade Triton X-114 in PBS to three cycles of phase separation. One cycle consists of two steps, one at 4° and the other at 37°, the latter yielding a detergent-rich lower phase consisting of 12% (v/v) Triton X-114 and a detergent-depleted upper phase, which is discarded. The detergent-rich phase is diluted to 2% (v/v) with fresh PBS before the next cycle. For large volumes, no centrifugation is required as phase separation will occur within 16 hr at 37°. After the third cycle, the detergent phase can be stored for years at -20°. ** M2-agarose can be conveniently packed in a used HiTrap column (Pharmacia). To this purpose, remove the red top and cut 2-mm plastic at the top of the column, using a razor blade. Remove the black plug and empty the column, taking care to save the filters. Empty HiTrap columns are not available. Alternatively, use any kind of column. * PMS is light sensitive. Always keep protected from light.
