A two-dimensional, solid-phase microextraction (micro SPE) capillary electrophoresis (CE) MS/MS method is used for protein complex characterization. The tryptic digest of a yeast ribosome complex containing 75 proteins is analyzed. Searching databases against the MS/MS data identifies 80-90% of proteins within the complex.

A solid-phase extraction capillary electrophoresis (CE)-MS/MS system is described which allows the automatic modulation of CE conditions during the analysis. Upon detection of an ion (or ions) above a pre-set threshold the voltage applied to the inlet of the CE system was increased to achieve 'peak parking' and facilitate the unambiguous assignment of a site of phosphorylation in endothelial nitric oxide synthase.

Localization of labile posttranslational modifications by electron capture dissociation: The case of gamma-carboxyglutamic acid

ECD FT ICR MS was applied to the localization of post-translational modifications. Sulfated peptides and peptides containing γ-carboxylated glutamic acid residues were analysed by ECD, IRMPD and CID. Only ECD gave the cleavage of the peptide backbone without the removal of the modification.
