Ectopic sprouty phenocopies loss of EGF-R signaling, and endogenous sprouty expression depends on EGF-R activity. Decreasing sprouty activity enhances the phenotype of dominant active EGF-R but not dominant active Ras or Raf. sprouty also inhibits signalling via the RTK Torso in the embryo. Biochemical and immunohistochemical studies in cultured cells show that Sprouty localizes to the inside of the plasma membrane and interacts with Drk and Gap1.

This paper reports the identification of a gene Dof which encodes a novel cytoplasmic protein with coiled-coil domain and ankyrin repeats. It is required in both mesoderm and trachea for their migration and for MAPK activation via FGF-R signaling in these cells. Whereas dominant active Breathless fails to rescue the effects of Dof mutants, dominant active Ras and Raf do. However, the effects of activated Torso and D EGF-R are independent of Dof. Dof therefore appears to act specifically downstream of FGF-Rs in the activation of Ras, and not other RTKs (see also [18••]).

This paper reports the identification of heartbroken, which is required for both mesodermal and tracheal migration and interacts genetically with heartless and breathless. Dominant active Ras but not dominant active Hearthless or Breathless rescues the heartbroken mutant phenotype indicating that heartbroken is downstream of these FGF-Rs and upstream of Ras. heartless- dependent activation of MAPK phosphorylation depends on heartbroken activity however heartbroken is not required for Dpp signaling in the mesoderm, or DEGF-R-dependent MAPK activation during tracheal morphogenesis or early embryogenesis. This suggests that hearthbroken acts specifically in FGF-R signaling and not signaling via other RTKs (see also [10••]).

The gene columbus was identified on the basis of its requirement for PGC migration towards the somatic gonad. columbus encodes the Drosophila homolog of HMG CoA reductase (hmgcr). hmgcr is expressed at high levels in the somatic gonad precursors. Ectopic expression of hmgcr can redirect germ cell migration to novel sites, suggesting that this gene is involved in the production of an attractive signal. All point mutants in hmgcr affecting germ- cell migration map to the catalytic domain, suggesting that enzymatic activity is essential for germ cell attraction.

Border cells are shown to migrate as a patch of cells which retain some of their epithelial polarity. DE-cadherin is required in both the border cells and the germline for their migration, and genetically reducing the amount of DE-cadherin slows the rate of border-cell migration.

Tissue-specific recombination is used to produce mutant clones in border cells. Females containing clones in which 2R was homozygous were screened for effects on border-cell migration. 6000 mutagenized lines were screened, and 20 mutants causing defects in migration were isolated. All but one of these was recessive homozygous lethal. Mutants were placed into 16 complementation groups, one of which corresponds to the known gene costal-2. The other complementation groups may represent new loci required for border-cell migration.
