Nature 1998, 394:494-498. EEA1 is found to possess two binding sites for the GTP-bound form of the endosomal GTPase Rab5, one at the amino terminus and one at the carboxyl terminus. The carboxy-terminal Rab5 binding site is juxtaposed to a PI(3)P binding FYVE zinc finger and the localisation of EEA1 to endosomes is regulated by PI3′-kinase and Rab5. EEA1 is found to be required for endosome fusion in vitro and overexpression of its carboxy-terminal part leads to an apparent fragmentation of endosomes in vivo. These findings might explain why wortmannin inhibits endosome fusion in vitro and why this inhibition is counteracted by the presence of excess Rab5-GTP.

An affinity isolation method results in the purification of 22 cytosolic proteins binding Rab5·GTP. One of these, EEA1, is found to be the only cytosolic component required to support endosome fusion in vitro. High concentrations of EEA1 strongly stimulate endosome fusion and obviate the need for PI(3)P and Rab5. A docking assay suggests that EEA1 acts prior to SNAREs at a docking step preceding endosome-endosome fusion.

Certain proteins are shown to be sorted to the lumen of the yeast vacuole. This sorting requires the PI(3)P 5-kinase Fab1p [44•,45•] and the products of vps class E genes. Studies of acidification-deficient yeast mutants show that fab1 and vps class E mutants lack internal vesicles within endosomes and vacuoles, suggesting that Fab1p and Vps class E proteins are involved in MVB formation.

EMBO J 1998, 17:4930-4942. Yeast mutants defective in endosome-to-lysosome transport exhibit increased levels of PI(3)P, suggesting that its turnover has been impaired. The mutants accumulate intralumenal vesicles within endosomes and vacuoles. It is proposed that PI(3)P degradation might involve its sorting into intralumenal vesicles that can be degraded by endosomal/vacuolar hydrolases.

Incubation of many cell types with the PI3′-kinase inhibitor wortmannin is known to cause the expansion of endocytic compartments. In a human melanoma line, this effect is shown to depend on endocytic membrane delivery and wortmannin treatment reduces the number of intralumenal vesicles within endosomes. This implicates PI3′-kinase in MVB morphogenesis.
