This study characterizes the physiological properties of individual RM neurons during, and in the absence of, anesthesia and provides the first evidence for the existence of OFF cells in the unanesthetized, normal rat. Nearly all of the recorded cells had state-dependent discharge, with ON cells most active during waking and OFF cells most active during slow-wave sleep.

Demonstrates that the behavioral hyperalgesia produced by intracolonic zymosan is attenuated by APV or L-NAME microinjected into the RM but is facilitated by microinjection of DNQX. None of the antagonists used had any effect on behavior in untreated animals. This finding provides evidence for simultaneous activation of pain inhibitory and facilitatory pathways in the presence of inflammation.

Given that excitation of the dorsal horn cells by iontophoretic application of excitatory amino acid agonists was not suppressed, the authors propose that the inhibition probably acts presynaptically, perhaps on primary afferent terminals.

Demonstrates that placing a rat's tail in hot water inhibits dorsal horn cell responses evoked by noxious heat applied to the hindpaw. At the same time, the motor response evoked by hindpaw heat changes from hindlimb flexion to hindlimb extension, without a change in motor latency. Therefore, modulatory effects on dorsal horn cells may be correlated with changes in withdrawal pattern rather than withdrawal latency.

Pseudorabies virus is a retrograde axonal tracer that can be used to trace multisynaptic connections; the number of synapses that are traversed depends on survival time. After short survival times, neurons in the ipsilateral ventral horn and intermediolateral cell column are retrogradely and transneuronally labeled by injection of pseudorabies virus into a tail withdrawal muscle. After longer survival times, labeled cells, which presumably represent interneurons involved in motor control, are found in the ventral, intermediate and dorsal gray, bilaterally. Spinal neurons labeled by the muscle injections were never retrogradely labeled after injection of cholera toxin B into midbrain and thalamic somatosensory regions. These results suggest that interneurons involved in somatic motor control are a distinct population from those involved in the rostral transmission of somatosensory information.
