To investigate the association between CpG methylation and gene silencing in vertebrates, proteins that cofractionated with the methyl-CpG binding protein, MeCP2, were isolated using Xenopus oocyte extracts. Sin3 and histone deacetylase - both of which are presumed to repress gene activity by modifying chromatin structure - were identified, suggesting that MeCP2 recruits the Sin3/histone deacetylase complex to promoters, thereby leading to gene silencing.

Dnmt1, the predominant maintenance and de novo DNA methyltransferase in mammals, undergoes alternative sex-specific splicing of the 5′ exons. Whereas the pachytene spermatocyte-specific exon interferes with translation of Dnmt1, the oocyte-specific exon produces an amino-terminal truncated protein that is highly expressed at later stages of oocyte growth. The oocyte-specific form of Dnmt1 is an excellent candidate for conferring the maternal-specific pattern of methylation.

Four of these offspring survived to maturity and proved to be fertile, indicating that the paternal-specific imprint is therefore established prior to meiosis in most spermatocytes.

The mechanism by which Igf2 is imprinted is here investigated using a series of mutations. Replacement of the endogenous H19 gene by the luciferase gene had no effect on Igf2 imprinting, indicating that the H19 RNA is not required. Although paternally-derived luciferase was expressed inconsistently, Igf2 expression was not affected, suggesting that the two genes are not regulated by a strict competition of the promoters for the H19 endodermal enhancers. Finally, the Igf2 paternal allele-specific methylation proposed to be important for Igf2 expression may not be required in embryos as Igf2 activation caused by the large H19 deletion is not disrupted in Dnmt1-deficient mice.

To assess the role of a 2 kb region of paternal-specific methylation that is located 2 kb upstream from the start of H19 transcription, a deletion was generated at the endogenous locus. As expected, when the mutation was paternally transmitted, H19 was activated and Igf2 expression was reduced. Surprisingly, maternal transmission of the deletion led to a reduction in H19 expression and activation of Igf2, indicating that this element positively influences H19 expression and/or inhibits Igf2 expression.

To test the hypothesis that the regulation of H19 and Igf2 imprinting is governed by a competition of the promoters for a common set of enhancers, mice that contained an extra set of enhancers and mice in which the enhancers were moved from their normal location were generated. Mice harboring a duplicated set of enhancers on the maternal chromosome expressed the maternal allele of Igf2. When the enhancers were moved from their normal location to a point between H19 and Igf2 on the maternally-derived chromosome, Igf2 was expressed, thereby reversing the direction of the imprint. The investigators conclude that H19 promoter strength alone is not sufficient to silence Igf2.

This publication reports evidence for two additional transcripts at the 3′ end of UBE3A: a downstream sense transcript that is maternally expressed and an antisense transcript that is paternally expressed. The authors suggest that the antisense transcript may serve to regulate the expression of the UBE3A gene.
