    • This review summarizes many recent results concerning the genesis of spontaneous activity by developing spinal networks in the chick embryo. It compares the type of activity produced in ovo and in vitro, emphasizing the existence of a 'fingerprint' in the pattern of muscle activity generated during spontaneous movements in ovo and in vitro. This fingerprint is a decrease in the probability of firing immediately after an intense burst. In vitro recordings have shown this to be attributable to shunting of the motoneuron membrane by depolarizing synaptic drive. Although indirect, this result suggests that the pattern of synaptic input to motoneurons in ovo is similar to that in vitro. The paper also summarizes the role of post-episode, network depression in the genesis of spontaneous activity by spinal networks. In addition, several possible functions of synchronized network activity are discussed.

    • A and glycinergic antagonists, bicuculline and strychnine, were used (instead of AP5, CNQX and the nicotinic cholinergic antagonists), the inter-episode interval lengthened and became erratic. Collectively, these results indicate that spontaneous activity produced by spinal networks is unlikely to depend on the details of network architecture. In addition, they show that networks can compensate for major disruptions of their connectivity and suggest that GABA/glycinergic and EAA/cholinergic networks may be regulated differentially in this respect.

Neuron. 20:1998;305-315. The authors used genetic manipulations to investigate the factors regulating synaptic efficacy at the neuromuscular junction of Drosophila. For this purpose, they overexpressed either a constitutively active catalytic subunit of PKA or a mutant form of the regulatory subunit with a reduced affinity for cAMP (which reduced PKA activity) in developing Drosophila muscle. Overexpression of the catalytic subunit decreased the average amplitude of spontaneous end-plate currents (quantal size), whereas overexpression of the mutant regulatory subunit had the opposite effect. The decrease in quantal size resulted in an increase in presynaptic glutamate release by an unknown mechanism. The authors propose that the increased presynaptic release involves a retrograde signal from muscle to nerve. The time course of the PKA-dependent changes in quantal size was investigated by increasing PKA activity in normal muscles by bath-application of a membrane-permeant form of cAMP. This procedure decreased quantal size within minutes in a concentration-dependent manner. It was shown that this decrease could be explained, in part, by a reduced postsynaptic conductance to iontophoretically applied glutamate.

    • Rutherford LC, Nelson SB, Turrigano GG BDNF has opposite effects on the quantal amplitude of pyramidal neuron and interneuron excitatory synapses. Neuron. 21:1998;521-530. Previous work by this group [44] identified the existence of activity-dependent scaling of quantal amplitudes in pharmacologically treated cortical cultures. In their earlier work, they demonstrated that chronically (48 h) increased activity depressed quantal amplitude, whereas decreased activity had the opposite effect. In this paper, the authors test the hypothesis that the neurotrophin BDNF mediates these effects. They found that application of BDNF to cultures containing TTX prevented the increase in the quantal amplitude of AMPAR-mediated EPSCs (from pyramidal cells to pyramidal cells) that would normally have occurred when activity was blocked. By contrast, application of BDNF to otherwise drug-free cultures increased the amplitude of AMPAR-mediated EPSCs from pyramidal cells onto GABAergic (inhibitory) bipolar cells. Application of the TrkB-IgG fusion protein, which binds endogenous BDNF and neurotrophin 4/5 (NT-4/5) with high affinity, increased AMPAR EPSCs in pyramidal cells but had no effect on those produced in bipolar cells. Collectively, these results suggest that the activity-induced scaling of AMPAR-mediated EPSC amplitude in these cultures is mediated (at least in part) by activity-dependent release of BDNF acting on TrkB receptors. Interestingly, the effective concentration of BDNF was much lower at pyramidal-pyramidal synapses than at those between pyramidal and bipolar cells. These differences are compatible with a model in which the release of BDNF is proportional to the level of network activity. At a high level of network activity, the correspondingly high level of released BDNF will selectively enhance pyramidal-bipolar synapses; in turn, this will increase the effective inhibition of pyramidal cell firing by bipolar cells, which will tend to return overall network activity to control levels. Therefore, BDNF has complex actions on circuit function that depend not only on its actions at particular synapses, but also on the way changes in synaptic efficacy affect the recruitment and ehavior of the network as a whole.

    • B receptors) only modestly increases burst duration and inter-burst intervals. The involvement of postsynaptic effects was rendered unlikely by two observations. First, blockade of AMPA receptor desensitization with cyclothiazide only slightly changed burst length. Second, the amplitude of current responses induced by brief applications of glutamate to distal apical dendrites showed no dependence on the time after a burst. Transmitter depletion was implicated in burst termination by two main observations: first, in the presence of 500 mM mannitol to increase glutamate release, a burst greatly depressed the frequency of spontaneous EPSCs; and second, drugs that reduced the probability of transmitter release (baclofen and adenosine) lengthened both the inter-burst interval and the burst length. Collectively, these findings suggest that population bursts in the hippocampal slice terminate when transmitter depletion at active terminals reduces the strength of recurrent excitation below a critical level. This mechanism for periodic activation of hippocampal networks is qualitatively similar to that proposed for developing spinal networks, although the precise mechanisms of the activity-dependent depression may differ between the two networks.

    • Direct participation of starburst amacrine cells in spontaneous rhythmic activities in the developing mammalian retina

    • •]. The work described in this paper investigated this proposal directly - although in a different species - by recording the activity of both cell types in the retina of the neonatal rabbit. Simultaneous whole-cell recordings were obtained from morphologically identified, displaced starburst amacrine cells (in the ganglion cell layer) and from ganglion cells. These recordings showed that both cell classes were synaptically active, thereby providing direct experimental evidence for the participation of starburst amacrine cells in retinal waves. Significantly, however, the frequency of activation of starburst and ganglion cells was similar, in contrast to the model predictions that starburst amacrine cell activity should be more frequent. The presence of rhythmic synaptic inputs to starburst amacrine cells raises the possibility that some other cell type - possibly GABAergic amacrine cells - could be involved in wave generation. Unfortunately, the pharmacology and reversal potential of the synaptic drive to the starburst amacrine cells was not established, so the identity of their synaptic inputs remains unknown. This study also established that starburst cells do not generate action potentials during their rhythmic synaptic drive, indicating that that some other mechanism, perhaps graded release of transmitter, must be involved if these cells are responsible for retinal waves.

The precise mechanisms responsible for the generation of retinal waves are unknown. Drawing on recent pharmacological experiments in the ferret retina [70], this paper describes a computational model to account for wave generation. The model attempts to explain the spatiotemporal dynamics of waves visualized using fura-2am, a calcium indicator, in ganglion cells of ferret retinas. It proposes that wave generation occurs in the amacrine layer, which is presynaptic to the ganglion cells and that the ganglion cells themselves do not participate directly in wave generation, acting instead as a 'readout layer'. Importantly, the ganglion cells are postulated to exhibit a distinct threshold for activation. The authors identify the existence of a post-wave refractory period, typically lasting 40-60 s, during which time previously active cells cannot participate in another wave. The authors provide arguments and data showing that wave dynamics and boundaries require a two-layer model and cannot be explained by a simple one-layer model in which cells are spontaneously active and exhibit a refractory period. Instead, the observed refractory periods require a two-layer model in which the functional refractory period of the ganglion cell layer arises from the interaction of the amacrine refractory period with the ganglion cell threshold.
