Puckey LH, Lawn RM, Knight BL. Polymorphisms in the apolipoprotein(a) gene and their relationship to allele size and plasma lipoprotein(a) concentration. Hum Mol Genet 1997; 6:1099-1107. A study of several polymorphisms in the apo(a) gene promoter in both Caucasian and Indian populations. The results indicate no direct association of any particular variant with Lp(a) level. Haplotype analysis provided evidence for groups of apo(a) alleles, each with a restricted range of sizes and associated with different Lp(a) concentrations. Linkage studies give some insight into how the apo(a) protein has expanded to produce the various isoforms.

The Apo(a) gene is the major determinant of variation in plasma Lp(a) levels in African Americans

Mooser V, Scheer D, Marcovina SM, Wang JP, Guerra R, Cohen J, Hobbs HH. The Apo(a) gene is the major determinant of variation in plasma Lp(a) levels in African Americans. Am J Hum Genet 1997; 61:402-417. The variation in Lp(a) concentrations in African Americans, as in Caucasians, was mainly attributable to sequence changes at or near the apo(a) gene locus. There was no evidence for a single high-expressing apo(a) allele to explain the high Lp(a) concentrations in these subjects.

J Clin Invest 1997; 100:1400-1405. A description of the effect of a mutation in the hepatic transcription factor HNF-4.
