By isolating gp96 from cells with and without functional TAP molecules, the authors of this paper demonstrate that peptides transported into the ER in a TAP-dependent or TAP-independent manner can associate with gp96. This study shows that the repertoire of gp96-associated peptides is indeed representative of the ER peptide pool and is not influenced by the MHC haplotype.

In addition, it is demonstrated that as little as 1 ng of gp96-peptide complex is able to induce a specific CTL response.
