A correction has to made for changes in the separation current with focus current Since the separation current decreases with increasing focus current, the rate of introduction of molecules to the detection region also decreases. However, in this experiment the separation current changed by only 10%, and the effect on molecules counted was not noticeable over the 25-s data set.

The signal-to-noise ratio of a peak was calculated by the formula (S - B)/ sqrt(B), where S is the peak height and B is the average background in the vicinity of the peak. The values used for the calculations in Figure 10A were as follows: S = 560 and B = 280 (analog); S = 53 and B = 3 (singlemolecule counting). The values used for the calculations in Figure 10B were as follows: S = 490 and B = 310 (analog); S = 40 and B = 3 (single-molecule counting).
