    • Production of aberrant promoter transcripts contributes to methylation and silencing of unlinked homologous promoters in trans
    • This paper provides the first indication that a transcriptional trans-silencing locus can act by producing aberrant transcripts of promoter sequences. This finding extends and confirms the widespread importance of the RNA-DNA interaction as a possible signal for de novo methylation in plants, as previously reported [6,7].

    • The authors found that premeiotically induced DNA methylation suppresses homologous recombination in the fungus Ascobolus immersus. This finding could have broad implications in the maintenance of eukaryotic genomes, in which DNA methylation associated with DNA repeats may contribute to inhibit recombination. In fact, homologous recombination between dispersed DNA repeats could lead to deleterious chromosomal rearrangements.

    • This paper describes a new homology-dependent gene silencing (HDGS) phenomenon in the diploid oomycete plant pathogen Phytophthora infestans. It was found that a silenced transgenic strain is able to impose transcriptional gene silencing on a wild-type strain in heterokaryosis, indicating that silencing is dominant and acts in trans. Interestingly, the presence of transgenes is not essential for maintaining the transcriptional silenced status. These findings sustain a model involving a trans-acting molecule that is capable of transferring a transcriptional silencing signal between nuclei.

    • Taming of transposable elements by homology-dependent gene silencing
    • Using the hybrid dysgenesis syndrome in Drosophila as a model, the authors demonstrated the existence of a homology-dependent gene silencing (HDGS) mechanism that represses the transpositional activity of the I element. This result demonstrates that transposable elements are prone to and can be tamed by HDGS, strongly supporting the theory that HDGS mechanisms are processes that may have emerged during the course of evolution as a defense system against these elements.
