The authors demonstrate convincingly that any homology of a viral genome with a silencing transgene, whether it is a viral gene or a plant gene, suppresses the replication of the virus. Also, the article is the first one to show that the 3′ part of the post-transcriptionally silenced gene is methylated more extensively than the 5′ part, suggesting that the process of transcription is involved in the generation of the silencing signal.

This interesting and well organized article contains results on different aspects of post-transcriptional gene silencing. Homology of the transgene to the 3′ part of the silenced genes produces more efficient silencing than does homology to the 5′ part to confer RNA-mediated virus resistance. 60 nucleotides of homology between the silenced transgenes and the target RNA are sufficient to mediate RNA degradation. The arrangement of the transgenes influences the strength of post-transcriptional gene silencing. Promoterless transgenes cannot induce virus resistance in transgenic plants. Last, but not least, virus-resistant lines all contain transgenes in which not the promoter but the transcribed region is methylated.

The switch appears to be associated with changes in the potential for expression of chitinase genes. Furthermore, the authors showed that, once silencing is established, the competence for silencing can persist in dormant as well as in actively growing and newly established shoot meristems.

Remarkably, a strict unequivocal and direct correlation between the quelling phenotype and the production of an unexpected sense-oriented chimeric transcript from the silencing locus is observed. Nevertheless, intended expression of promoter-driven sense or antisense RNA did not increase quelling.
