This study demonstrates the effectiveness of direct in vivo gene transfer of soluble receptors of interleukin-1 and tumor necrosis factor-α to the knees of rabbits with antigen-induced arthritis. Moreover this study shows that anti-arthritic effects were also seen in contralateral control knees, suggesting that sustained local inhibition of disease activity in one joint might confer an anti-arthritic effect on other joints.

J Orthop Res 1998, 16:330-339. This study is a successful BMP-2 gene therapy in large segmental bone defects in the rats using the ex vivo strategy. This paper further demonstrates the morphological difference in the pattern of bone formation between the gene transferred defects and those treated with recombinant human BMP-2, suggesting the importance of continuous expression of BMP-2 at the site of repair for the proper formation of bony trabecular architecture.
