Nature 1997, 389:498-501. This study provides the first real evidence that the BSE agent causes both BSE and 'new variant' (v) CJD. It does this by identifying the BSE 'signature' of incubation periods and pathology in mice after transmission from brain of a case of vCJD, but not after the transmission of other types of CJD.

The in vitro limited conversion of normal human protease-sensitive PrP (called PrPSEN by these authors) by the protease-resistant forms of PrP from scrapie and BSE into additional protease-resistant PrP (called PrPRES) suggests that both scrapie and BSE may not be readily transmissible to humans. It serves to highlight the effect of species barriers on transmission or host range control.

This study on PrP glycoform patterns in different strains of scrapie, including the BSE-derived 301V, shows that even strains from the same origin can have significant glycoform differences. This is exemplified by the strains 79A and 263K, which are at opposite ends of the glycoform spectrum and yet are both derived from the SSBP/1 isolate of scrapie. It is concluded that strains of transmissible spongiform encephalopathy with similar glycoform profiles may not necessarily have a common source.

The major gene controlling mouse scrapie incubation is Sinc (also known as Prni), Gene targeting was used to construct mice in which the endogenous Prnp allele was modified to express PrPB instead of PrPA. PrPA and PrPB are PrP variants arising from codon 108L/F and 189T/V dimorphisms (where the letters represent in single-letter code the amino acids encoded by these codons). Incubation periods were markedly reduced when these mice were challenged with a mouse-passaged BSE strain, demonstrating that PrP dimorphisms at codons 108 and 189 control incubation time and that Sinc/Prni and Prnp are congruent.
