LIN-2 and LIN-7 are functionally required for localization of the LET-23 receptor to the basolaleral membrane domain of the vulval precursor cells. LIN-7 may act directly in receptor localization as LIN-7 binds to LET-23 in vitro.

These binding interactions may mediate the localization of nNOS to the synapse and suggest a role tor PSD-95 in nNOS signaling.

This important paper presents data suggesting that chapsyn-110 can cluster N-methyl-D-aspartate (NMDA) receptors and Shaker channels. Chapsyn-110 can form a multivalent protein complex by binding to PSD-95, and this protein complex can bind and thereby cluster the Shaker channel and NMDA receptor. The authors show that either the Shaker channel or the NMDA receptor can cluster with chapsyn-110 when each is co-expressed with chapsyn-110 in heterologous cells.

Presents the identification of GRIP, which has seven PDZ domains and can bind to the carboxyl terminus of subunits in the AMPA receptor at excitatory neurosynapses. Overexpression of the carboxyl terminus of a subunit of the AMPA receptor prevents its localization to the synapse, suggesting that GRIP may function in receptor localization.

An outstanding paper showing that InaD is involved in localizing the transient receptor potential (TRP) channel to the rhabdomere and acts as a scaffold for signaling proteins in the photoreceptor cell in the fly. The authors isolate two new InaD alleles using an impressive genetic screen InaD has five PDZ domains, and three of these PDZ domains bind to different signaling molecules, namely, phospholipase C, TRP channel and protein kinase C.
