The authors find that the chimera moves toward the plus direction and conclude that it is the position of the motor domain which determines the polarity of the kinesin molecule. However, closer inspection of the ncd motor - Nkin stalk chimera reveals that the construct actually consists of the neck and catalytic domain of ncd fused to the neck and tail/stalk region of Nkin. The presence of the two neck regions directing movement in opposite directions accounts for the decreased velocity of this construct as a plus-end-directed motor. These studies show that future attempts to dissect the directional preference of kinesins will have to take the neck region into account.
